A multifuctional nanoplatform for drug targeted delivery based on radiation-engineered nanogels
2020
Abstract Under a rational design, combining biologically active molecules, ligands to specific cell receptors and fluorescent, radioactive or paramagnetic labels into a single nano-object can bridge the unique properties of the individual components and improve conventional sensing, imaging and therapeutic efficacies. The validation of these functional nano-objects requires careful testing both in terms of physico-chemical properties and biological behaviour in vitro and in vivo, prior to translation into the clinic. Ionising radiation of aqueous polymer solutions is a viable strategy to produce multifunctional nanogels from aqueous solutions of hydrophilic polymers. By proper selection of …
Synthesis and characterization of a new insulin conjugated nanogel for biomedical application
2015
Insulin, a metabolic hormone involved in glucose metabolism, plays also a neuroprotective role in the central nervous system being able to revert the cytotoxic processes induced by Aβ42,a peptide involved in Alzheimer's disease. To reach the brain insulin have to across the BBB therefore an additional delivery strategy results to be necessary. For these aim we performed an insulin conjugated nanogels (NGs-In). Nanogels (NGs) have a great potential in the development of “smart” nanocarriers for (bio)molecular drugs and contrast agent for bioimaging. They are formed by physically or chemically crosslinked polymer networks, characterized by a large and flexible surface available for multivalen…
Polymerization reactions and modifications of polymers by ionizing radiation
2020
International audience; Ionizing radiation has become the most effective way to modify natural and synthetic polymers through crosslinking, degradation, and graft polymerization. This review will include an in-depth analysis of radiation chemistry mechanisms and the kinetics of the radiation-induced C-centered free radical, anion, and cation polymerization, and grafting. It also presents sections on radiation modifications of synthetic and natural polymers. For decades, low linear energy transfer (LLET) ionizing radiation, such as gamma rays, X-rays, and up to 10 MeV electron beams, has been the primary tool to produce many products through polymerization reactions. Photons and electrons in…
Nose-to-brain delivery of insulin enhanced by a nanogel carrier.
2018
Recent evidences suggest that insulin delivery to the brain can be an important pharmacological therapy for some neurodegenerative pathologies, including Alzheimer disease (AD). Due to the presence of the Blood Brain Barrier, a suitable carrier and an appropriate route of administration are required to increase the efficacy and safety of the treatment. Here, poly(N-vinyl pyrrolidone)-based nanogels (NG), synthetized by e-beam irradiation, alone and with covalently attached insulin (NG-In) were characterized for biocompatibility and brain delivery features in a mouse model. Preliminarily, the biodistribution of the "empty" nanocarrier after intraperitoneal (i.p.) injection was investigated b…
Micro- to Nanoscale Bio-Hybrid Hydrogels Engineered by Ionizing Radiation
2021
Bio-hybrid hydrogels consist of a water-swollen hydrophilic polymer network encapsulating or conjugating single biomolecules, or larger and more complex biological constructs like whole cells. By modulating at least one dimension of the hydrogel system at the micro- or nanoscale, the activity of the biological component can be extremely upgraded with clear advantages for the development of therapeutic or diagnostic micro- and nano-devices. Gamma or e-beam irradiation of polymers allow a good control of the chemistry at the micro-/nanoscale with minimal recourse to toxic reactants and solvents. Another potential advantage is to obtain simultaneous sterilization when the absorbed doses are wi…
Radiation-Engineered Multi-Functional Nanogels As Nanoscale Building Blocks Of Useful Biomedical Devices
2011
Radiation-engineered multifunctional nanogels: a versatile platform with theranostic capabilities for cancer
2018
Under a rational desing, combining biologically active molecules, ligand to specific cell receptors and fluorescent, radioactive or paramagnetic labels into a single nano-object can bride the unique properties of the individual components and impruve conventional sensing, imaging and therapeutic efficaces.
Numerical simulations of the kinetics of pulsed electron beam induced synthesis of nanogels in dilute aqueous polymer solutions
2018
Nanogel can be produced by irradiation of dilute aqueous polymer solutions. The nanogels are formed primarily through intramolecular radical-radical crosslinking reactions between two polymer radical sites.
On the nature of macroradicals formed upon radiolysis of aqueous poly(N-vinylpyrrolidone) solutions
2020
In this work we have explored the nature of macroradicals formed upon radiolysis of aqueous poly(N-vinylpyrrolidone) (PVP) solutions using pulse radiolysis, density functional theory (DFT) and literature data. On the basis of literature data on site-specific kinetics of hydrogen abstraction from simple amides and spectra corresponding to specific radical sites on the same amides we have assessed the distribution of H-atom abstraction by `OH radicals from different positions on the pyrrolidone ring and the polymer backbone. Pulse radiolysis experiments performed at different doses per pulse and different PVP concentrations demonstrate that the H-abstracting radiolysis products are not quanti…
